Use the EM_CHARFROMPOS message:

        if( LineIndex > -1 )
            // ...
    }

    void __fastcall TForm1::Memo1Click(TObject *Sender)

        int LineIndex = Memo1->Perform(EM_EXLINEFROMCHAR, 0,
Memo1->SelStart);
        if( LineIndex > -1 )
            // ...
    }
