Sugars & HPLC - post column derivatization?

Hello,

I 've got to setup a system for quantitative measurement of glucose,
fructose, saccharose and may be polyols (sorbitol, mannitol) using HPLC.

I've got two options:

1. Using ionexclusion chromatography on a cation exchanger like Biorad's
AMINEX HPX 87 H with diluted sulfuric acid as mobile phase

or

2. Using a silica based amino-propyl phase with ACN/Water (75/25) as mobile
phase.

Detection limit should be at least 100 ng Gluc or Fruc absolute. As
detector I'll have to use an RI, which I'm afraid is not sensitiv enough.
In particular when mobile phase is ACN/Water. Electrochemical detectors are
not available and not affordable, respectively.

I've read about post column derivatization using Thymol in concentrated
sulphurous acid, or Periodic reagent. Unfortunately I don't have access to
more detailed information about exact procedures.

Any hints and reccomendations are wellcome.

Thank you very much in advance!

Rainer Hofmann
