Try 

   \mathcode`,="xyzz

where

   x is the "part of speech" (you probably want 6, for punctuation)
   y is the family (0 for the "roman", ie un-bold, un-italic etc, family)
   zz is the character within the font, in hex (probably the ascii for ",")
