How to create transparency clipart

Dear all,

    I'm so interesting to create clipart by myself. But I found it is
quite difficult to create a transparency clipart.

  What I used to do is to get selection of image by magic wand, then
paste it to a new blank image with transparency background.

   But it will turn to colored background when I save it to other file
format and insert in other application like Powerpoint or Word.

  Does it require any special skill? Be grateful to learn from you.
