I'm from Hamilton originally and have crossed to and from Buffalo many times.
As you know, July1-4 is a holiday weekend for both Americans and Canadians, so
you can definitely expect significantly increased times to cross. It depends on
what time of day you're crossing: two hour delays at heavy times are not un-
common.

To get to the 401 from Buffalo you have to travel on some secondary roads. I
believe the most direct route would to take the Queen Elizabeth Way (QEW -
the only major highway on the Cdn. side) west heading to Toronto. After you
pass Hamilton (ugly view of steel mills) you want to look for Highway 6 North,
which will take you directly to the 401. Then head west. Highway 6 is an 80
km/hr road vs. the QEW and 401 which are 100 km roads so, again depending on
traffic, it shouldn't take you too long.
