Briefly, I have been working with a particular fern for a while now and have reached a point where I could use some advice from anyone experienced with plant tissue culture, somatic hybridization, etc. I'd like to discuss this by e-mail since the questions I have may not be of general interest. If anyone out there would like to talk about this, here follows an outline of the problem. We would like to do some work on a mutation in the gametophyte generation which renders them entirely female. This is someone else's mutant and our proposed study requires further mutation of spores carrying the gene for that trait. Being male infertile makes a simple self-cross impossible. A possible question is: Can haploid protoplasts from the same species potentially be fused and regenerated to make a homozygous, spore-producing sporophyte? Please respond if you feel you can help. A simple yes or no from someone in the know regarding the above question would be a tremendous help. Thanks, -- Mitch Day daymitc@cwis.isu.edu
