Whenever I measure CD spectra of protein, must I have to measure CD spectra of standard(eg. saccharose 1%, saccharose 5%) for baseline correction or measure once per year or so? Thanks. Lee, Ji Hyun.

Yes, you should measure a blank for each set of buffer conditions and cell pathlength. You should also calibrate the machine with camphorsulfonic acid every few weeks or so.
