Position available immediately for an ambitious post-doc to join new
lab investigating the role of the cytoskeleton in ion channel
regulation by tyrosine phosphorylation. Experience in protein
biochemistry desired but not required. Some experience with
electrophysiological techniques a plus.
Our previous findings suggest the intriguing hypothesis that tyrosine
phosphorylation dependent suppression of the potassium channel Kv1.2
occurs by a mechanism that involves the cytoskeleton. Our current
efforts are directed at testing this hypothesis. To accomplish this, we
use a combination of biochemical and electrophysiological techniques,
along with channel mutagenesis, to determine the regions within Kv1.2
that participate in both channel-cytoskeleton interactions as well as
in channel suppression. A particular strength of approach is that it
allows us to draw conclusions regarding the causal relationships that
exist between biochemical and physiological events. Our research has
the potential to establish, for the fist time, a detailed molecular
model of Kv1.2 suppression, identify a new paradigm for channel
modulation (i.e. tyrosine phosphorylation dependent channel
interactions with the cytoskeleton), and provide the basis for new
avenues of research into the regulation of other ion channels.
